Where should I start? Does the bass/treb/sub on the head unit be all at 0? Where should I start at the amp Gain/ bass?

If you start with your amp gain at 0 you will have nothing from the amp. Turn your volume up to about 2x your normal listening level. Start with everything around 2. Then adjust your highs, then mids, then bass. I adjust mine to where I start getting some distortion then back them off 1/2 - 1 full adjustment level... For a good sounding system, you want just enough bass not to drown out your mids or highs.
